بررسی تأثیر استفاده از aml-dc های آلوژنیک در القاء ctl اختصاصی و بررسی فعالیت ctl های ایجاد شده به صورت ex-vivo

چکیده
در روش معمول تولید ctl جمعیت سلول های t اتولوگ سه بار توسط dc های لوکمیک آلوژن همان بیمار تحریک می شوند. اما در روش پیشنهادی این تحقیق t سلهای بیمار aml دو بار توسط dc های لوسمیک اتولوگ و یک بار توسط dc های لوکمیک آلوژن بیمار دیگر تحریک شدند و از بلاست های اتولوگ، سلول های اتولوگ نرمال و رده اختصاصی سلول های هدف نشاندار شده از طریق آزمون سیتوتوکسیسیتی محاسبه گردید. توان dc های لوسمیک در mlr آلوژن بطورمعناداری بیش از سلول های قبل و بعد از کشت بود. سیتوتوکسیسیته ضد لوسمیک ctl های القا شده توسط dc های لوسمیک بیش از سلول های t اتولوگ تیمار نشده، سلول های t تیمار شده با il-2، سلو های تیمار شده با il-2 و تحریک شده با بلاست های تازه و نیز بلاست های کشت شده تیمارنشده با سیتوکین بود. بنظر می رسد پروتوکول جدید اتولوگ آلوژن با تولید ctl های ضدسرانی قوی تر می تواند بعنوان استراتژی موفقی در ایمنوتراپی بیماران all مطرح شود.
